卵成熟におけるサイクリンB翻訳開始機構の分子形態学的解析

| Research Abstract |
サイクリンBmRNAの翻訳開始に関わる細胞質ポリアデニル化エレメント結合蛋白質(CPEB)とPumilioの機能を詳細に知るため、アフリカツメガエル卵成熟過程におけるこれら蛋白質のリン酸化、蛋白質間相互作用、及びサイクリンB翻訳との関わりを調べた。その結果、1)卵成熟過程でPumilioはCPEBとサイクリンBmRNAへの結合部位であるPufドメインではなく、これらとの結合に関与しないN末部でリン酸化される、2)卵成熟過程でCPEBのリン酸化時期とサイクリンBの翻訳開始時期が一致する、3)CPEBの予想リン酸化部位三個所を変異させるとPumilioとの解離が阻害される、4)このリン酸化部位変異CPEBの強制発現は卵成熟に影響を与えない、5)CPEBは予想リン酸化部位が存在しないC末部でPumilioと結合することが明らかになった。
また、PumilioやCPEBと共に、翻訳開始に関与すると考えられる他の蛋白質の卵成熟への関わりを探り、同一切片を用いたin situハイブリダイゼーション法と免疫組織化学法により、Maskin、Pumilio、切断・ポリアデニル化因子(CPSF)、ポリA結合蛋白質(PABP)、CPEB、ポリA合成酵素(PAP)、真核生物開始因子4E(eIF4E)が、卵核胞の内部及び周囲に同所的に局在することを示した。さらに免疫沈降法により、1)Maskin-PAP、Pumilio-CPEB、CPEB-eIF4Eの結合が卵成熟過程で外れる、2)Pumilio-eIF4E、CPSF-eIF4Eの結合が蛋白質間の結合からRNAを介した結合に変化する、3)CPSF-PAP、PABP-PAPの結合が新たに生じることを示した。
以上の結果から、卵成熟過程におけるサイクリンB1の翻訳調節機構を考察した。
