| Research Abstract |
我々が自己免疫性腸症関連自己抗原として単離したautoimmune enteropathy-related 75 kDa antigen (AIE-75)は,小腸・腎近位尿細管の刷子縁および内耳上皮細胞に発現している.消化管上皮におけるAIE-75の発現は,絨毛側に強くcrypt側には認めない.従って,AIE-75の発現が細胞周期の影響下にあることが考えられる.今回,AIE-75の発現制御機構を解明する目的で,AIE-75遺伝子の上流領域の転写活性を検討した,まずAIE-75の発現に関与するシス領域を同定する目的で、21エクソンからなるAIE-75遺伝子の全長とその上流領域約10kbを含むPACクローンを得た。種々の解析よりAIE-75遺伝子の翻訳開始点は転写開始点から+97bp,プロモーター領域は転写開始点の上流203bpと推測された.そこで,ルシフェラーゼアッセイ用のレポーターベクターpGL3basicにプロモータ領域(-203)から翻訳開始点(+97)までをルシフェラーゼ遺伝子とin-frameとなるように組み込んだconstruct (pGL-300)を作製した.さらに,転写開始点より上流-121bpから転写開始点を含むpGL-212,およびpGL-212のうち-63から-9までの54塩基対を欠く(pGL-212δ)コンストラクトを作製し,これをNIH3T3細胞およびAIE-75の発現が確認されている大腸癌細胞株KM12に形質導入し,ルシフェラーゼアッセイを行った.その結果,転写活性はPGL-300>pGL-212δ>pGL-212の順に強いことがわかった.このpGL-212δの欠失部にはSP1結合モチーフの他に転写制御因子GCF結合モチーフと細胞周期依存性転写制御因子HiNF-C結合モチーフが含まれており,これらによる発現制御を受けている可能性が示唆された.
