Research Abstract

To produce anti-swine NK receptor antibody, we immunized swine lymphcytes or membranous antigen to mice and rats. However, the antibodies could not stained swine NK cells. We sometimes tried the same experiments, but antibodies for staining swine NK cells were not produced. On the other hand, we tried swine small intestine transplantation. At the first time, the experiments were in failure, however gradually the experiments became in success. Finally, we could have long survival cases after amall intestinal transplantation. Next we injected α-Gal Cer to the small bowel recipient pigs. However bcause α-Gal Cer were injected to few transplantated pigs, we could not compare the survival period between injected group and no injected group. We can not confirmed relationship between α-Gal Cer and NKT cells in pigs, because we could not produce swine NK antibodies. However these results don't deny hypothesis that α-Gal Cer induce immunological tolerance after small intestinal transplantation. NKT cells are well recognized in mice and human. We found relationships between NKT cells and immunological tolerance in human living related donor liver transplantations. And we have experienced many murine experiments about NKT cells. It may be necessary to examine the method for administration of the drugs. To confirm induction of immunological tolerance using NKT cells and α-Gal Cer, we need further investigation in murine intestinal transplantation model.
