| Research Abstract |
1)ビーグル犬の胃壁に5×5cm大欠損部を作り、同部の再生案験を行った。シリコンシートで内腔面から隔離した欠損部に再生の足場となるコラーゲンスポンジを埋殖した。平滑節の再生を促すために、骨髄よりの間葉系幹細胞に加えて、消化管の平滑筋細胞を含漬させる実験を行った。さらに、細胞外マトリックスにFGF、EGF等組織増殖因子を複合化すると再生を促進することが判明した。
2)小腸と大腸は各々5cmの置換実験をビーグル犬で行った。これまでの予備実験で平滑筋の再生と術後の収縮を防ぐことが、臨床応用に向けた大きな課題であることが明らかになっていたので、今回はまずは生体内分解性ステントを用いて再生の場を保持させ、筋細胞の源となる細胞を自己の平滑筋から導入した。
3)総胆管(CBD)はVater乳頭から1cmの位置で1cm長の置換実験を行った。ビーグル犬10匹へ埋入した。至適なステントの抜去時期を決定するための埋入実験を継続している。
