マイクロアレイCGH法を用いた原子爆弾放射線の継世代的影響調査

Research Abstract

この科研補助による研究において、アレイCGH法が有効に使用されるために必要な基礎的要求;(1)大きな欠失を確実に検出するための実験条件の確立、(2)コンピュータ画像処理による、ゲノムコピー数の増減の迅速なる同定、(3)膨大なデータを処理するためのコンピュータソフトの開発とその改良は、前年度にほぼ達成された。本法により、放射線により生ずる100万塩基対(1Mb)を超えるような大きな欠失型変異が見逃されることなく、しかも迅速に検出されることが明らかになった。我々は、約120万塩基対(1.2Mb)毎の間隔で常染色体に位置する約2400種類のBac-クローンを選び、それらを3枚のガラスに分けてスポットした。このマイクロアレイを用いて、被爆者の子供40名および対照群の子供40名から得たゲノムDNAを試料とした集団調査を昨年度来続けている。その結果、2名の対象者で変異型が検出された。この対象者の両親を調べたところ、これらの変異は少なくとも片親から遺伝していることが分かった。従って、新規突然変異は現在のところ同定されていない。この集団調査とは別に、放射線照射細胞より得たゲノムをこのアレイを用いて解析したところ、欠失型変異が数ヶ所に検出された。これをモデルとして、リアルタイムPCR法、サザン法による、変異部分の大きさを同定する実験条件を確立した。従って、集団調査の過程で検出された変異の意味付けが迅速に実施可能となった。対象集団を拡大して本法による検査を行うことにより、従来はモデル動物でしか明らかにされていなかった、放射線の継世代的影響(遺伝的影響)が、ヒトにおいて、初めて明らかになる可能性が現実のものと成った。また同時にこの集団調査の過程で得られる多型変畢は、それらの有する生物学的影響を明らかにする、格好な機会を提供するものと考えられる。
