| Research Abstract |
当該年度においては、昨年度調製したtabacco ringspot virus(TRSV)、arabis mosaic virus(ArMV)及びchicory yellow mottle virus(CYMV)のサテライトRNAのマイナス鎖由来ヘアピン型リボザイム遺伝子を用い、PRV mRNAに対するリボザイムの設計を行った。また、これらのリボザイムの触媒ドメインに存在する切断活性を持つために必要な塩基配列のうち、1塩基を他の塩基に置換したコントロール遺伝子も合わせて調製した。このコントロール遺伝子は、リボザイム遺伝子と同一の塩基長であり、気質結合ドメインは同一の配列になっていることから、リボザイムの切断活性を除いたアンチセンスRNAをコードしている。これらのコンストラクトを大腸菌内にクローニングしたが、多数のコンストラクトの調製を行ったため、クローニングに多大な時間を費やしてしまい、細胞内でのウイルス抑制効果を検討できなかった。今後、一連の遺伝子がクローニングできたら、細胞内での効果を検討していきたいと考えている。
ところで近年、遺伝子の発現抑制にRNA干渉(RNAi)が効果的との報告もあり、このRNAiは標的RNAと結合する塩基長は二十数塩基長であるが、リボザイム及びこのコントロールRNAについても十数塩基長であることから、リボザイム及びこのコントロールRNAもRNAi様の効果を示している可能性も否定できないと考えられる。従って、PRVに対するRNAi研究も並行して研究を進めていく予定である。
