Research Abstract |
We examined the kinetics of mature dendritic cells (DC) in the lung and the subsequent P6 protein-specific immune response after intratracheal challenge of outer membrane protein of non-typable Haemophilus infleunzae (NTHi) in a mice. A rapid increase of matured DC was found in the pulmonary lymph nodes (LNs) after challenge of NTHi OMP. These mature DC peaked at Day 1 and gradually declined until Day 7. The in vitro P6-specific LN cell proliferation suggested the P6-specific antigen presentation to the naive T cells through the mature DCs in the pulmonary egional LNs. A strong link between the P6-specific antigen presentation and the P6-specific antibody production in the BAL fluids as well in sera was demonstrated. The P6-specific Ig production in sera might be, in part, explained by the emergence of P6-specific IgG and IgM forming cells in spleen. The P6-specific IgA production in BAL fluids was associated the plasma cell proliferation beneath the bronchial epithelium and the localization of IgA. These data demonstrated that the influx of matured DC into the pulmonary regional LN could link with the development of adaptive immunity in the lung as well as systemic circulation, and suggested pivotal role of DC in airway immunization by NTHi OMP.
