Research Abstract	ラット新生仔心室筋培養細胞を用いた実験受容体レベルでAngIIを阻害し(アンジオテンシンII受容体拮抗薬RNH-6270による前処置による)、CT-1を投与した際にSTAT3活性化が無処置群と比較して抑制されていた。リガンドレベルでAngIIを抑制し(アンジオテンシノジェンに対するアンチセンス処置群と培養液交換後にアンジオテンシン変換酵素阻害薬テモカプリルの活性体であるテモカプリラート前処置を行う)、CT-1を投与した際にSTAT3活性化が無処置群と比較して抑制された。リガンドレベルでのAngIIの抑制処置を行い、閾値下濃度の外因性AngIIを前投与しCT-1によるSTAT3の活性化がおこることを見出した。(AngII抑制下での外因性AngIIのSTAT3活性化の濃度依存性を検討し、「閾値」を決定した後に施行した。) カベオラドメインの機能的阻害(filipinの投与)を行うとCT-1によるSTAT3の活性化が抑制された。また、filipinを除去した際の(filipinの効果は可逆的であると報告されている)にはその抑制が解除された。上記、それぞれの条件下での心筋肥大の程度を検討した。CT-1による心肥大は(1)RNH-6270、(2)テモカプリラート+アンジオテンシノジェンのアンチセンス処置、(3)filipinの投与によりそれぞれ抑制された。カベオラとAngII受容体およびgp130の会合をウェスタンブロット法により確認した。
