Research Abstract |
We have done a fundamental research on the development of a new technology of direct hemoperfusion aiming to remove immunosuppressive cytokines from the blood of cancer-bearing hosts. Immunosuppressive substance adsorption (ISA) fiber was prepared by chemical modification of a polystyrene-based composite fiber. In brief, the composite fiber is treated with N-methylol-α-chloroacetamide in a mixture of sulfuric acid and nitrobenzene in the presence of paraformaldehyde. Sterile cylindrical plastic columns with a capacity of 2 ml were prepared by filling 0.4 g of ISA fiber. Tumor-bearing rats were made by implanting KDH-8 cells (hepatocelllar carcinoma) subcutaneously into the back of WKAH/Hm rats. Direct hemoperfusion of tumor-bearing rats using these columns significantly reduced the tumor growth and. prolonged the survival of them. These columns efficiently removed TGFb, VEGF, and IL-6 from the blood of tumor-bearing rats.
