子宮内膜の発癌過程におけるコネキシン機能異常のメカニズム

Research Abstract	子宮内膜癌、子宮内膜増殖症におけるE-cadherin遺伝子のメチル化の検討 connexin(Cx)の機能発現と細胞接着のkey proteinであるE-cadherinは密接な関連があることが期待されるが、これらの蛋白の遺伝子異常、発現異常、細胞内局在の変化がどのように細胞の増殖や分化、発癌過程で関わり合っているかは不明な点が多い。本研究では子宮内膜癌104例、子宮内膜増殖症17例、正常子宮内膜21例より抽出したDNAを用いE-cadherin遺伝子のプロモーター領域のメチル化を検索した。また、E-cadherinとCx26、Cx32の発現と局在を免疫組織化学的に確認した。子宮内膜癌細胞を用いてこれらの遺伝子の発現と細胞間コミュニケーション(GJIC)を計測した。培養細胞ではCxが正常に発現している場合、E-cadherinの発現の有無によってGJICの強さは決定された。さらに、E-cadherinの発現が減弱していた細胞はいずれもメチル化が観察された。また、癌組織では正常内膜、内膜増殖症ではメチル化は観察されないが、癌では分化度が下がるほどその割合が高くなった。一方、Cxの発現・局在異常は内膜癌はもとより内膜増殖症においても高頻度に観察された。E-cadherinの発現とGJICは密接に関わっているものの、GJICの減弱は子宮内膜の発癌過程のearly eventとしておこり、E-cadherinの発現の減弱は遺伝子のメチル化を介してlate eventとして起こるものと考えられた。
