子宮内膜症の進展および子宮筋腫の発症とダイオキシン受容体関連蛋白多型との相関

Research Abstract	ホルモン感受性の遺伝子群の特定を、ダイオキシン関連遺伝子群を中心に進めている。ダイオキシン関連遺伝子群の検討に関しては、ダイオキシン核内移行因子2の解析をデータベースおよび過去の試料から分析を行っているところである。また、遺伝子データベースを用いた解析により、ダイオキシンレスポンシブエレメントが関連している遺伝子群のスクリーニングを行っている。一定の遺伝子に関連して変動する遺伝子群を検索する方法を開発しているところであるが、この方法を、ダイオキシンレスポンシブエレメントに応用し、関連する遺伝子群の特定を進めている。今後、ダイオキシンのみではなく、性ホルモンに関連して動く遺伝子群に応用していく予定である。エストロゲン、プロゲステロン、アンドロゲンといった性ホルモン群を調節している部位と強く相関する遺伝子群を特定し、子宮内膜症、子宮筋腫の病巣における発現、子宮内膜症、子宮筋腫の発生と、治療予後との関連性について検討していく予定である。また、子宮の発生に大きく関わるHOX遺伝子の変異解析パネルを作成した。HOX遺伝子群は組織の形態形成に本質的に関わるホメオティック遺伝子群であるが、子宮形成においてもHOX13が関わるばかりでなく、着床期の子宮内膜における発現、子宮内膜症の有無による子宮内膜における発現量の変化が指摘されている。新たな、ホルモン感受性遺伝子の検索を進めるとともに、HOX遺伝子群およびダイオキシン関連遺伝子群の子宮内膜症組織および子宮筋腫組織における発現解析も同時に進行させていく予定である。
