Research Abstract |
Extraction of BMP in large scale, selection of scaffold, cartilage induction on a membrane material and osteoinduction in the host body by re-implant of induced cartilage were experimented. Extraction of BMP is on the line in stable, and at least 1000mg of BMP is obtained per month. Considering BSE problem, starting material of BMP was changed to pig bone from bovine bone. There was no difference between pig and bovine BMP in the ability of bone induction. Selection of polymers showing biological function both in vivo and in vitro was performed. Candidates of polymers were attelo collagen, gelatin, agarose, poly-saccharide, PLA-PLGA co-polymer, polyehylmethacrylate and polytetrafluoroethylene. Almost all polymers showed excellent property as BMP carriers, eventually, polytetrafluoroethylene was selected as a membrane forming material. PLA-PLGA co-polymer, polyehylmethacrylate and methylcellulose will be important material for this research project in future. Porous titanium was also investigated, and showed that osteoprogenitor cells were well harvested in the titanium pore. Cartilage formation was occurred on the polytetrafluoroethylene membrane by inducing factor of BMP. This Cartilage system was implanted into subcutaneous tissue of rat, which cause the osteoinduction at 3 weeks after implantation. The objective of this project was then achieved by this result.
