Research Abstract

TERT (telomerase reverse transcriptase) is acknowledged as the rate-limiting determinant of telomerase and transcriptional regulation of the TERT gene plays a key role in the activation of telomerase. The hamster and human TERT promoters share common protein binding sites, such as E-boxes or GC-boxes which are for Myc and Sp1 respectively. Recent studies have demonstrated that expression of human TERT (hTERT) is affected by Sp1 but not c-Myc in many cases,
To study the effect of transcriptional factor Sp1 on the transactivation of TERT, we investigated the expression of Sp1 and TERT mRNAs with RT-PCR during carcinogenesis of the hamster tongue induced by DMBA and tried to make insight into the relationship between their expressions.
The expression of hamster TERT mRNA was well correlated with the telomerase activity. Coincidentally, we found that the expression of Sp1 and hamster TERT were upregulated in dysplasia and squamous cell carcinoma tissues and they were closely linked with each other. These results suggest that Sp1 is a crucial regulatory factor for the hamster TERT transactivation during the carcinogenesis of the tongue.
