Research Abstract	カルボニル還元酵素発現によるマウス卵巣がん細胞の性状変化の解析昨年度、マウスカルボニル還元酵素(CR)の発現ベクターを作製し、細胞にトランスフェクトして、CRタンパク質の発現を調べたが、低かったので、今年度、新たな発現ベクターに変更し、これにCR cDNAを挿入し、CR発現ベクターを作製した。これをマウス卵巣がん細胞にリポフェクトアミンを用いて導入した。トランスフェクトした細胞を10%ウシ胎児血清を含むDMEM培地で培養した。細胞数はMTT法に準ずる方法で測定し、細胞形態は位相差顕微鏡で観察した。CRと血管内皮細胞増殖因子(VEGF)の発現をウエスタンブロット法で検討した。本発現ベクターにより卵巣がん細胞でCRの強い発現を認めた。CR発現に伴い、細胞増殖や細胞形態に明らかな変化を認めなかったが、VEGFの発現は低下することが明らかになった。来年度以降、本細胞系を用いてCRによるVEGFの発現抑制の機序、CRを発現させたがん細胞では転移が抑制されるかなどを検討する予定である。
