| Research Abstract |
1)MT-4細胞でのウイルス抗原陽性細胞の推移:MT-4細胞にHHV-6B(Z29株)を感染させ、HHV-6の前初期蛋白に対する抗体IEA/ex3、後期蛋白に対する抗体OHV-3を用いてウイルス抗原陽性細胞数を経時的に観察した。その結果、IEA/ex3抗原陽性細胞は感染後4時間目から陽性細胞が出現し、48から72時間目には約60%の細胞が抗原陽性となった。また、OHV-3抗原陽性細胞は24時間目から抗原陽性細胞が出現し72時間目に約50%の細胞が抗原陽性となった。
2)MT-4細胞におけるHHV-6増殖曲線作成:MT-4細胞にHHV-6B(Z29株)をMOI=1で感染させ、ウイルス増殖を調べた結果、新生HHV-6粒子の効率的な産生が確認された。
3)リアルタイムPCRによるウイルスDNA量の推移:2)と同様にウイルス接種後のMT-4細胞を経時的に採取し、DNA抽出。採取された検体中のHHV-6DNA量をリアルタイムPCR法で解析した。2)の結果を裏付けるように、HHV-6DNAは感染後徐々に増加し、感染後72時間でピークに達しその後徐々に低下してゆくことが確認された。
4)HHV-6感染MT-4細胞の細胞死についての検討:MT-4細胞にHHV-6B(Z29株)を感染後、24時間、48時間後に細胞を採取し、IEA/ex3抗体、アネキシン抗体を用いてフローサイトメーターで陽性細胞の検討を行った。その結果、感染48時間後の細胞で、HHV-6抗原陽性細胞だけでなく、HHV-6抗原陰性細胞も高率にアポトーシスに陥っていることが確認された。
