Research Abstract

ヒト・マトリックスメタロプロテイナーゼ-7(MMP7)は高い蛋白質分解活性をもち、癌転移に関与することが知られている。本酵素の阻害物質は有効な癌治療薬になる可能がある。pH5-9の広いpH域に特徴的な最適pHを有している。本酵素の反応機構は不明である。我々が行なってきたこれまでの研究から、酸性側の活性解離基としてGlu198またはZnルイス酸、塩基性側の活性解離基としてTyr216やTyr219などの可能性が考えられた。今回、塩基性側の活性解離基の同定と役割を解明することを目的とした。部位特異的変異導入により、活性部位に存在する3個のTyr残基(Tyr193,Tyr216,Tyr219)をそれぞれPheに変換した。野生型酵素では、酸性側pKa(pK_1)は4.4、塩基性側pKa(pK_2)は9.8である。Tyr193PheとTyr216Pheは、野生型とほぼ同じpK_1とpK_2値を示した。一方、Tyr219Pheでは、それぞれ5.7と9.4となり、pK_1が1.3ユニットも塩基性側にシフトした。いずれの変異酵素も最適pHでの酵素活性はほぼ同程度であり、以上の変異導入は固有の酵素活性には影響を及ぼさないことが示された。上で示されたpKaのシフトは、Tyr219が塩基性側の活性解離基ではないこと、またその水酸基が塩基性側解離基に直接的な影響を及ぼさないこと、ただし、Tyr219の水酸基により酸性側解離基はプロトン解離が促進されることが示された。塩基性側解離基の同定は未解明のまま残されたが、反応機構の解析において特異で貴重な知見が得られた。
