| Research Abstract |
本年度は昨年度ミシガン大学で行ってきた内耳遺伝子導入を当院の研究室で行えるよう実験機材の購入(手術道具、ポンプ他)及びreporter gene(GFP、lacZ)を組み込んだvirusを購入して内耳への遣伝子の組み込んだvirusがおこなうことが出来るように主にセットアップを行った。そして実際にモルモットを用いてreporter gene(GFP、lacZ)を組み込んだadenovirus及びretrovirusの導入を行いgene expressionが起こっていることを確認した。Virusは直接蝸牛の中央階に投与する方法と鼓室階に投与する方法でgene expressionの生じている部位を確認した。Adenovirusの投与ではgene expressionの起こる範囲は中央階投与では蝸牛内全体に起こった。また鼓室階投与では鼓室階のfibroblastにgene expressionが生じた。またRetrovirusでは中央階投与ではfibroblast及び血管条の細胞にgene expressionを認めた。また鼓室階投与ではわずかにfibroblastにgene expressionみとめた。免疫応答に関しての実験モデルはvirusを鼓室階に投与して検討している。はじめにgene expressionの起こっている期間を免疫抑制状態(サイクロフォスファマイド投与群)とコントロールで比較した。次に初回のvirus投与後3週間あけてvirusを2回投与行い免疫応答による細胞傷害をcytochochleogramで観察しているところである。さらに抗T cell抗体で免疫染色するために標本を作成している。
