Research Abstract

光学活性ジホスフィンと1,2-ジアミンをともに配位子とする塩化ルテニウム(II)錯体は,2-プロパノール中,塩基の存在下,様々な単純ケトン類の不斉水素化反応において極めて高い活性と立体選択性を示す。平成17年度は、塩化ルテニウム錯体を水素化ホウ素ナトリウムで処理して得られるRuH(η^1-BH_4)(diphosphine)(diamine)が塩基無添加条件でケトン類の水素化に高い活性を示すことを見出した。さらにRuH(η^1-BH_4)(diphosphine)(α-picolylamine)がt-アルキルケトン類の不斉水素化還元に有効であることを発見した。これまで嵩高いケトン類の不斉還元における鏡像体過剰率は不十分であったが、この問題を解決する新しい手法を提供できたといえる。さらに、反応機構の詳細な検討も行い、我々が以前から提唱している、ケトン基質の水素化遷移状態における金属-配位子二官能性機構を支持する結果を得た。
また、核酸、糖医薬、液晶材料等の合成において有用な原料となるβ-ヒドロキシニトロアルカンやβ-アミノニトロアルカンの効率的な合成を可能にする、全く新しい環境調和型非金属触媒(アミノ有機ボラン-水型クラスター触媒)の開発にも成功した。これまでほとんど皆無であった、水素結合のみを推進力とする触媒的炭素-炭素結合形成手法の実現である。また本触媒は複数の酸と塩基が協同的に働く多機能性をもつが、同時に高機能性をも示し、ニトロアルドール反応のみならず直戯アルドール反応やアルコールのアセチル化反応にも適用できることが明らかになった。
さらに、独自に設計・開発したロジウム錯体が、芳香環やヘテロ芳香環の炭素-水素結合を直接アリール化する優れた触媒となることも見出した。有機化学における基本的かつ汎用性の高い反応となる可能性を秘めている。
