Research Abstract	我々が同定した新規視床下部LPXRFaペプチドであるGnIH(鳥類LPXRFaペプチド)は鳥類の脳下垂体に作用してゴナドトロピンの放出を抑制することを明らかにした。またfGRP(両生類LPXRFaペプチド)は両生類の脳下垂体に作用して成長ホルモンの放出を促進することを明らかにした。次に、GnIH遺伝子とfGRP遺伝子をクローニングした結果、GnIH遺伝子はGnIHと2種の遺伝子関連ペプチド(GnIH関連ペプチド)を、fGRP遺伝子はfGRPと3種の遺伝子関連ペプチド(fGRP関連ペプチド)を、それぞれコードしていることがわかった。これらの遺伝子関連ペプチドの作用を解析して、fGRP関連ペプチド(fGRP-RP2)はプロラクチンと成長ホルモンの放出を促進することを明らかにした。さらに、魚類LPXRFaペプチド(gf LPXRFaペプチド)の作用を解析したところ、この新規視床下部ペプチドは魚類のゴナドトロピン放出を促進することが見いだされた。魚類LPXRFaペプチド遺伝子にも複数の関連ペプチドがコードされており、これらの関連ペプチドにも同様の作用があることが示された。本研究では、さらに以上の新規視床下部LPXRFaペプチドによる脳下垂体ホルモン放出制御機構を明らかにするために、新規視床下部LPXRFaペプチド受容体の同定を試みた。その結果、鳥類からGnIH受容体のクローニングに成功した。クローニングしたGnIH受容体を培養細胞に強制発現させてペプチド結合実験を行い、GnIH受容体であることを証明した。
