Research Abstract

最近、我々は鳥類のウズラの脳から、ゴナドトロピンの放出を抑制するゴナドトロピン放出抑制ホルモン(GnIH)を発見した。本年度はミヤマシトドなど他の鳥類からもGnIHを同定して、GnIHは鳥類の生殖を制御する重要な脳ホルモンであることを明らかにした[主論文J.Endocrinology 182:33-42]。次に、GnIHの作用機構を明らかにするために、GnIH受容体を同定した。GnUH受容体は7つの膜貫通領域を持つ新規のGタンパク共役型受容体であり、GnIHと特異的に結合した。GnIH受容体は脳下垂体に発現しており、GnIHは脳下垂体のGnIH受容体を介してゴナドトロピンの放出を抑制することが解明された[主論文J.Endocrinology 184:257-266]。さらに、GnIHの発現制御機構を解析して、松果体および眼球から分泌されるメラトニンがGnIHニューロンに局在するメラトニン受容体を介してGnIHの発現を誘導することを明らかにした[主論文Proc.Natl.Acad.Sci.USA 102:3052-3057;プレスリリース:米国 The Scientistなど]。この成果はNature Reviews Neuroscience 4月号に紹介される。
一方、GnIHはC-末端にLPXRFa構造を持つ特徴があることから、GnIHの同族ペプチド(LPXRFaペプチド)を他の脊椎動物からも同定して機能を明らかにした。両生類のLPXRFaペプチド(fGRPとその遺伝子関連ペプチド)は成長ホルモンとプロラクチンの放出を促進させ、魚類のLPXRFaペプチドはゴナドトロピンの放出を促進することを明らかにした。また、LPXRFaペプチドの起源を明らかにするために、最も原始的な脊椎動物である無顎類ヤツメウナギから同族のペプチドを探索した。
