新規視床下部ホルモンによる脳下垂体ホルモン合成・放出の新しい制御機構

Research Abstract

従来、脊椎動物の生殖は脳の視床下部ニューロンが合成する生殖腺刺激ホルモン放出ホルモン(GnRH)に支配されていると考えられてきた。GnRHは脳下垂体から生殖腺刺激ホルモンを放出させる作用がある。一方、生殖腺刺激ホルモンの放出を抑制する脳ホルモンは存在しないと長く考えられていたが、我々は鳥類のウズラの脳から生殖腺刺激ホルモン放出抑制ホルモン(GnIH)を発見した。本年度はGnIHが生殖腺刺激ホルモンの放出・合成を抑えて、生殖腺の発達と機能維持を抑制することを明らかにした(Endocrinology147:1187-1194;Brain Res.1050:94-100)。また、他の鳥類からもGnIHを同定して、GnIHは鳥類の生殖を制御する重要な脳ホルモンであることを明らかにした(J.Endocrinol. 182:33-42;Horm.Behav.49:550-555)。さらに、新規のGタンパク共役型受容体であるGnIH受容体を同定して、GnIHの作用機構を明らかにした(J.Endocrinol.184:257-266)。また、メラトニンがGnIHの発現を誘導することを明らかにした(Proc.Natl.Acad.Sci.USA 102:3052-3057;Nature Reviews Highlight [Nat.Rev.Neuresci.6:264-265])。本年度は、さらに哺乳類、両生類、魚類の脳からGnIHと構造の類似する同族ペプチドを同定し、GnIHとGnIH同族ペプチドが広く脊椎動物の生殖制御に関わっていることを明らかにした(Proc.Natl.Acad.Sci.USA 103:2410-2415;J.Endocrinol.188:417-423;FEBS Journal, in press)。
