光合成アンテナをモデルとした超分子ナノデバイスの創製

Research Abstract

緑色光合成細菌の膜外アンテナ部=クロロゾーム中では、クロロゾーム型クロロフィル分子が自己会合することによって、アンテナ色素部を形成している。そのような天然型クロロフィルを模した様々な置換基を有する会合体モデル色素分子として、配位しやすい水酸基と水素結合能のあるカルボニル基とを有する環状テトラピロール類の配位性金属(Mg,Zn,Cd等)錯体を、天然産のクロロフィルを有機合成化学的手法で改変して調製し、その自己会合体の構造と機能を解析した。一方、天然のクロロゾーム型クロロフィルには、様々な部位がメチル化された分子が見られるが、どれも既存の有機合成法では導入しにくい。そこで、緑色光合成細菌の培養条件を変更することで、選択的なメチル化体の入手をまず試みた。嫌気性の緑色硫黄細菌の一種であるChlorobium vibiroformeでは、成育に必須な硫化物イオンの培地内での濃度を増大させると、12^1位のメチル化が抑制されることが判った。これを利用して、これまで天然界では検出されたことが無かった8位にプロピル基・12位にメチル基を有するバクテリオクロロフィル-cの単離に成功した。
次に、クロリン環の20位のメチル化は、色素分子の単量体のみならず、自己会合体の光学的な性質に対して大きな影響を与え、生理学的にも重要な因子となっている。しかしながら、有機化学的に20位のメチル化を行うことは難しく、生体酵素を利用することにした。Chlorobium tepidumの20位メチル化酵素(BchU)を、遺伝子操作によって大腸菌内で大量発現させ、単離精製することに成功した。様々なクロロフィル誘導体を基質にして、S-アデノシルメチオニンをメチル基供与体に用いて、BchUによるメチル化反応を行い、1段階で選択的に20位メチル化体を入手できた。
