Research Abstract |
A new breeding strategy using DNA marker in apple and pear was studied on the basis of the information from the saturated linkage maps efficiently and simultaneously constructed by these intergeneric hybrids. 1. The linkage maps of 'Fuji' apple and 'Oharabeni' pear were constructed with these 62 symmetric intergeneric hybrids lived by embryo rescue. The saturated map of 'Fuji' consisted of 17 linkage groups and 195 loci spanning 1348.1 cM, and that of 'Oharabeni' consisted of 17 linkage groups and 240 loci spanning 1345.4 cM, respectively. Each co-dominant locus of SSR and some genes were arrange with same order and genetic distance in each linkage groups of both cultivars, suggesting the high synteny between apple and pear genome. 2. About 350 EST clones obtained from the cDNA library in the young fruits of 'Osa Nijisseiki', 'Oharabeni' and 'Fuji' were sequenced and searched homology in BLAST. Most of the EST clones expressed in pear fruit showed high homology and single nucleotide polymorphism (SNP) with those in apple. Moreover, mutation of more than 5 base-pair insertion or deletion was found in the 6% of pear EST clones compared to the registered apple clones. 3. Some fruit characteristics such as fruit maturity, fruit shape, fruit weight, sugar and acid contents in the hybrids between 'Osa Nijisseiki' and 'Oharabeni' were investigated and mapped by QTL analyses. 4. The inheritance of the traits of columnar habit (Co), red leaf (Rl), red skin (rfs) and red fruit fresh in the progenies between 'Fuji' and 'Maypole' apple were analyzed. R1 and Co genes were mapped in the LG 9 and LG 10 of 'Maypole', respectively.
