Research Abstract

Lactoferrin (Lf) is an iron-binding protein present in colostrums, saliva, and intestinal mucos. The protein has a role in iron absorption and is an important antimicrobial component of the host defence. We hypothesized that supplemental Lf improves acute inflammation in endotoxin shock in neonatal calves. Newborn calves (10 d of age) were assigned to receive saline, Lf (200 mg/kg), or Lf (400 mg/kg). The calves were fed saline or Lf at 0900 and 2100 h. At 0900 h on next day, the calves received an i.v. injection lipopolysaccharide (50 ng/kg, E.coli 055 ; Shigma Chemical Co., St.Louis, MO) as endotoxin. In the calves treated with saline, plasma concentrations of ferrum, retinol, and lipoprotein fraction (VLDL and LDL) were decreased between 24 and 96 h after endotoxin administration. In the calves treated with Lf, these metabolites were not decreased by endotoxin injection. In addition, Lf feeding inhibited fever and production of haptpglobin, tumor necrosis factor-alpha (TNF), interleukin-6 (IL-6) following endotoxin treatment in calves. In summary, we have demonstrated that oral treatment of calves with Lf diminished the intensity of endotoxin-induced acute inflammation which was associated with significantly decreased acute phase protein and pro-inflammatory cytokines such as TNF and IL-6. These findings are in accordance with other results on anti-inflammatory properties of Lf in various in vitro models.
