蛋白工学的に新規作製したHGF-MSPキメラ(MFI)を用いた肝再生治療法の開発

Research Abstract

目的:HGF-MSPキメラ(MF1)の肝組織修復における有効性と安全性について検討した。方法:HGFとMSP遺伝子をリンカーでつなぎMF1遺伝子を作成、さらにレンチウイルスベクターシステムを用いてMF1蛋白を得た。In vitroの解析として、マウス肝細胞を用いて、MF1, HGFを培養液に添加し、増殖活性、抗アポトーシス作用を解析した。浸潤能の解析は肝癌細胞株HepG2、Huh-1を用いてmatrigel invasion assayを行った。in vivoの実験として急性四塩化炭素モデルにMF1, HGFを投与し、経時的に肝臓を採取し肝細胞増殖能、アポトーシスを検討した。さらにMF-1, HGF遺伝子導入した肝癌細胞の転移モデルを用い癌転移数を検討した。結果:in vitroではMF1の肝細胞増殖活性は、HGFに比べ若干弱いながらも強い活性が認められた。また抗アポトーシス作用はMF1でHGFとほぼ同様の効果を示した。また肝癌細胞はHGFで浸潤能が増強されたがMF-1は対照と同等であり、MF1は浸潤能を誘導しないことが確認された。In vivoで急性肝障害モデルでMF1投与によりALTの低下、肝障害の軽減が認められ、効果はHGF群とほぼ同様であった。また肝細胞分裂能はMF1投与で増加しTUNEL陽性細胞は優位に低下しておりin vitroを反映した結果であった。肝癌転移モデルではHGFが高率に転移を起こすのに対してMF1は対照群と同様であった。結論:HGF-MSPキメラ(MF1)は肝臓に対して増殖能、抗アポトーシス作用を有し、一方肝癌細胞の浸潤能は示さなかったことから安全かつ有用な組織修復因子となりうる可能性が示された。
