| Research Abstract |
EBVはバーキットリンパ腫から単離された170kbの二重鎖DNAウイルスである。Rajiはバーキットリンパ腫から1963年に樹立され、世界中の研究室で広く使われている細胞株のひとつである。50-60copy/cellのEBVゲノムを持ち、一部はintegrationしている。inverse-PCR法を用いてウイルス組み込み部位をclonihgした。BamHI-W, BamHI-D fragment上に組み込み部位は存在した。Host genomeとBamHI-Wとは組み込み部位において高いhomologyを示した。Chromosome 6q上にあるB-cellのdifferentiationに関与する遺伝子Bach2のintronにEBVの組み込み部位は存在した。Rajiにおいて同遺伝子の発現は認められなかった。同遺伝子は癌抑制遺伝子である可能性を示す報告もあり、RajiにおいてはEBVの同部位への組み込みが発がんに関与した可能性も考えられる。実際に9例のBurkitt lymphoma cell line,174例の臨床例を用いてBACH2の発現を調べた。Bach2はfollicular center lymphomaに高頻度に発現を認めMALTでは頻度は低かった。
(IB4)はKieff Eによって樹立されたlymphoblastoid cell lineであり、latent geneの解析にしばしば用いられる。IB4は4-5 copy/cellのEBVゲノムがhost genomeにintegrationしていると考えられている。われわれは、inverse-PCR法を用いてウイルス組み込み部位をcloningした。組み込み部位はoriP近傍のBamHI-C fragment, EcoRI-I fragment上に存在し、相同性は低かった。組み込み部位はchromosome 4q25に存在し、お互いに約6.5kbp離れていた。同部位は100kbp以上にわたりrepetitive sequenceが存在するが構造遺伝子は存在しなかった。
NAB-2はNorth American Burkitt tumorから樹立されたcell lineで1copy/cellのEBVがゲノムintegrationしている。CloningしたところEBVはREL, BCL11Aという発癌遺伝子のごく近傍にintegrationを認めた。現在integrationによるこれら発癌遺伝子への影響を解析中である。
