肝細胞癌治療におけるアポトーシス誘導、分子標的治療併用の試み-Transgenic mouseを用いた検討-

Research Abstract	肝癌培養細胞(PLC/PRF/5,Huh7,HepG2)に対し、まず PPARγ agonistとして使用可能なrosiglitazone, pioglitazoneを加え、3種類の細胞ともapoptosiswo誘導する事を見い出した。生理的な濃度でさらに抗癌作用を高めるために、抗癌剤との併用でapoptosisの促進が得られないかどうかを検討したところ、5FU, CDDP, Epirubicinの3種類の抗癌剤の内でEpirubicinでさらに有意にapoptosisが促進される事を見い出した。これらは臨床でも応用可能な組み合わせであり、今後in vivoでの併用効果を検討する。EGF receptorを介した細胞増殖系とPPARγの系はPPARγのligandにより、reactive oxygenを介してSrcからEGF receptorが活性化されることが示されてきており、最近EGF-receptorの阻害剤であるZD1839はPPARγ agonistによるExtracellular signal regulated kinase(Erk)の活性化を阻害する事が示された(JBC:278:46261-69)。よって我々が当初計画したPPARγ agonistとZD1839の併用は同時併用は単にPPARγ agonistの作用を打ち消すだけになる可能性が有る。よって、まずは、この系が培養細胞でも再現されるかどうかを検討し、細胞へ順序を変え、また併用して細胞増殖抑制効果をみる。 ZD1893は導入に時間がかかったが、その濃度依存性とtime courseを培養細胞で検討した結果、肝癌細胞(PLC/PRF/5,Huh7,HepG2)では、IC50は灼1μMであることが判明した。今後はこの両者(ZD1839,pioglitazoneまたはrosiglitazone)の相互作用をin vitroで検証し、nude mouse移植系でin vivoの検討を行なう。
