Research Abstract

Peroxisome proliferator-activated receptor(PPAR) is a member of the nuclear receptor family transcription factors. Three PPAR subtypes are known at present, PPARα,PPARδ, and PPARγ. Among these PPAR subtypes, PPARγ is expressed in the adipose tissues at a high level and involved in the regulation of adipocyte differentiation. Gastrointestinal epithelial cells also express PPARγ at a significant level and reports suggested the role of PAPRγ in the regulation of cell proliferation and cell death. However it is not well understood whether PPARγ plays a role in the protection of gastrointestinal mucosa.
Trefoil factor family(TFF) is a group of secretary peptides critically involved in the defense and repair of gastrointestinal mucosa. We examined the effect of PPARγ activation on the expression of TFF in gastric epithelial cells, and found that, among three TFF subtypes (TFF1,TFF2,and TFF3), PPARγ activation leads to the up-regulation of TFF2 expression. By detailed promoter analysis, we found the presence of a functional peroxisome-proliferator responsive element(PPRE) within the promoter sequence of the human TFF2 gene. Thus, the results of the present study suggest that human TFF2 gene is a direct target of PPARγ, and that PPARγ is involved in the defense of gastric mucosa through the regulation of TFF2 expression
