| Research Abstract |
フィラデルフィア陽性慢性骨髄性白血病(Ph+CML)はその類似疾患に慢性好中球性白血病(CNL)Ph陽性血小板増多症(Ph+ET)があるが表現型の違いが分子生物学的な違いによるかははまったくわかっていない。しかし、bcr-abl融合遺伝子のサブタイプの違いによりこの病型が決定されるのではないかと推測されている。この分子生物学的な違いは切断点部位の違いによるbcr遺伝子内アミノ酸配列部位の長短である。P230Bcr/AblとP210Bcr/Ablでのアミノ酸の違いは180個におよぶアミノ酸の違いでありこの配列の中にcalcium/phospholipid binding domain(CalB),Gap-Rac domain等機能を持つdomainがある。またb3-a2タイプP210Bcr/Ablはb2-a2タイプP210Bcr/Ablより疎水性βシートペプチドを構成している25アミノ酸があり3次構造が変化することがヒト病型に変化をもたらすと考えられている。以上のことからP230Bcr/Abl、b3-a2、b2-a2タイプP210Bcr/Ablをそれぞれ発現したトランスジェニック(Tg)マウス作成によりその表現型の違いがヒト臨床病型に側しているかを明らかにすることを目的として研究を行った。
P230Bcr/Abl Tgマウスを作成しそのマウスの表現型がヒト慢性好中球性白血病(CNL)やPh陽性血小板増多症(Ph+ET)に類似していることを確認し雑誌Bloodに発表した。
b3-a2、b2-a2タイプP210Bcr/Abl Tgマウスを作成に成功した。現在その病型を末梢血、臓器病理標本で確認中である。
