Research Abstract |
The function and expression of RP105 on B cells in autoimmune diseases have not been fully clarified, especially in human. To investigate the RP105 expression of autoantibody producing B cell subsets in patients with Systemic Lupus Erythematosus(SLE), we performed phenotypic and cell viability study for RP105-negative B cells in SLE. Expression of B cell markers on 105-negative and RP105-positive B cells was analyzed by a flow cytometry. The peripheral blood mononuclear cells were cultured with various stimulators with or without their blockers and were analyzed for expression RP105 on B cells. RP105-negative B cells form SLE showed increased expression of surface levels of CD38 and CD27 was found in RP105-negative B cells compared with RP105-positive B cells from SLE. In the culture with activated T cell and BAFF/APRIL, increased survival of RP105-negative B cells were found in SLE. In SLE, B cells, differntially express various markers of B cells and the survival was maintained by T cell stimulation and BAFF/APRIL stimulation. RP105-negative B cells may be targets for the treatment of SLE.
