Research Abstract	1.遺伝子導入による制御に関する検討遺伝子導入に関してはInvitrogenのシステムを用い新たに遺伝子導入実験を開始できる状況である。またすでにSTAT6の供与をうけており、発現実験は終了しin vivoの実験を開始するところまで来ている。肝阻血再灌流障害の制御に関しては、内因性に障害を制御する機構としてIL-18に注目しその発現と抗体投与による障害への関与を検討した。その結果IL-18は抗炎症性サイトカインの発現を抑制し障害を増悪させることが判明しHEPATOLOGYにpublishした。またノックアウトマウスを用い遠隔臓器(肺)における障害への関与も検討した。 2.Ischemic preconditioningに関しては、至適時間が決定できこれにより明らかに長時間の阻血再灌流障害が軽減されるという結果が得られた(p<0.01)。さらに組織学的好中球集積やchemokineの発現がreal-time PCRによる検討によりmRNAレベルで有意に抑制されるという結果が得られた。さらに阻血中に発現するmediatorに関してはmacrophage migratory inhibitory factorの発現が再灌流後経時的に免疫組織科学染色並びにmRNAレベルで認められた。 3,閉塞性黄疸肝の易感染性に関しては、肝切除後のTLR4の発現を検討し正常肝での感染と臓器障害との関連を証明し現在論文投稿中である。黄疸肝に関してもcytokine, chemokineの発現と好中球集積と臓器不全との関係に関してはすでに検討が終わり、現在易感染性の機序の解明を転写因子の発現やその後の蛋白発現より検討中である。
