Research Abstract |
We previously reported that immobilized phosphoprpteins and insoluble dentin were able to induce to mineral formation in metastable solution. However, it were not reported pH change that immobilized phosphoprotein induce apatite formation. In this study, phosphoproteins were crosslinked to an agarose beads and measured pH at several time in metastable solution. We investigated that usefulness of evaluation using electrical engineering in mineral induction. Phosvitin was used as a model phosphoprotein in this study. Phosvitin was immobilized to agarose beads. Three mineralizing solutions were used. Ten microliters of each sample were incubated in 3ml of metastable solutions at 37℃ in a shaker. Samples were measured pH at several time points and filtered. Then, samples were analyzed for calcium by atomic absorption spectrometry. Phosvitin crosslinked agarose beads were observed fall of pH 2.5h, 3h, 5h after beginning of incubation in the three solution. At the same time, Samples increased in analysis for calcium absorption spectrometry. These results suggested this method was more simple and precision.
