Research Abstract

1. To deduce the contractile. mechanism of Noradrenaline and adrenaline on porcine lingual arterial smooth muscle, we investigated changes in isometric tension and intracellular calcium level ([Ca^<2+>]_i) simultaneously. We assessed the relationship between the isometric tension and [Ca^<2+>]_i induced by 10^<-7>, 10^<-6>, 10^<-5>, 10^<-4>, 10^<-3>, 0.1, 0.5, 1.0, 2.0, 4.0, 6.0, 10.0, 20.0uM NA and Ad.
As results,
(1) NA and Ad increased the contraction and [Ca^<2+>]_i in a concentration-dependent manner.
(2) NA and Ad induced transient increase in [Ca^<2+>]_i followed by sustained levels, while they induced rapid and sustained contractions.
(3) In the contraction by NA Ad, phasic contraction depends on the release of Ca^<2+> from the intracellular Ca^<2+> store and tonic contraction depends on the influx of extracellular Ca^<2+>
2. To deduce the effects of Lidocaine on lingual, pulmonary and coronary arterial smooth muscle, we measured simultaneously the isometric contraction and [Ca^<2+> … More ]_i induced by 0.5, 1.0, 10.0, 20.0, 50μg/ml lidocaine after 40mM KCl, 5μM NA and Ad administered
As results,
(1) 1.0μg/ml lidocaine induced increases in tension and [Ca^<2+>]_i when 40mM KCl, 5μM NA and Ad were administered to lingual and pulmonary arterial smooth muscle.
(2) In coronary artery, the administration of 5μM NA and Ad induced no response and when 40mM KCl was administered, 1.0μg/ml lidocaine induced increases in tension and [Ca^<2+>]_i.
3. To deduce the effects of mepivacaine on lingual, pulmonary and coronary arterial smooth muscle, we measured simultaneously the isometric contraction and [Ca^<2+>]_i induced by 0.4μM, 4.0μM, 40μM, 0.4mM, 4.0mM mepivacaine after 40mM KCl administered.
As results,
(1) In lingual and coronary arterial smooth muscle, 4μM mepivacaine induced increase in tension and [Ca^<2+>]_i and 40μM, 0.4mM, 4.0mM mepivacaine induced decrease in tension and [Ca^<2+>]_i.
(2) In pulmonary arterial smooth muscle, 0.4 and 4μM mepivacaine induced increase in tension and [Ca^<2+>]_i and 40μM, 0.4mM, 4.0mM mepivacaine induced decrease in these. Less
