薬物依存形成に関与する分子群の簡便な検索法の確立および応用

Research Abstract

メタンフェタミンは我が国において最も乱用されている依存性薬物である。しかし、メタンフェタミンが薬物依存を引き起こす機構については、ほとんど分かっていなかった。小生は、平成12年度からこれまで、文科省科学技術振興調整費による研究助成を受け、『依存性薬物により誘発される精神障害の機構の解明の研究』をテーマに研究班を組織し、薬物依存に関する研究を行ってきた。これまでの研究によって本研究班から、薬物依存関連遺伝子としてtumore necrosis factor-α(TNF-α)およびtissue plasminogen activatorを見出した。しかし、それらの遺伝子を見つけるためには、依存モデル動物を用いて、脳内での発現変化や行動薬理学的な検討を行わなければならず、多くの薬物依存関連遺伝子候補タンパクをスクリーニングすることは、時間的にも費用面においても不可能に近い。当萌芽研究ではその問題を解決するため、培養細胞を用いて簡易に薬物依存関連遺伝子候補タンパクをスクリーニングできる薬物依存モデルの確立を目指している。
メタンフェタミンが薬物依存を引き起こす作用機序として、細胞間隙でのドパミン遊離量が増大し、精神毒性が引き起こされることが報告されていることから、メタンフェタミン添加によってドパミン遊離の増大が観察される細胞系を確立することができれば、薬物依存関連遺伝子のスクリーニングに活用することが出来ると考えられる。そこで、本年度は、PC12細胞を用いて検討を行ったところ、メタンフェタミンを3時間作用させた培養上清中のドパミン遊離量が、メタンフェタミンを作用させていないものに比べ有意に増加することが明らかとなった。さらに、薬物依存抑制作用を示すTNF-αをメタンフェタミンを加える1日前から作用させておくと培養上清中のドパミン遊離量が有意に抑制されることが明らかとなった。次年度は、TNF-α以外のこれまでに報告されている薬物依存関連遺伝子をPC12に作用させ、その効果を調べる予定である。
