Outline of Annual Research Achievements

Vector Controlのため、時に環境基準や野生動物への影響を度外視した散布がしばしば実施されている。ヒト健康を尊守する上で必要な処置であるが、この薬剤散布がどのような環境影響、野生動物への影響、更にはヒトへの影響があるのか、評価する必要がある。一方、Vector ControlにはDDTのみでなく、同時に様々なピレスロイド系農薬が使用されているが、この共散布が希少生態系にどのような影響を与えるのか、未だ報告はほとんど無い。更に、genome情報が乏しく、in vivo実験が困難な希少野生動物にこれら薬剤がどのような影響を与えるのか、それを評価するための方法も今だ確立されていない。以上の背景の基、当該研究では「Vector Controlで使用される薬剤のヒトおよび希少野生動物への毒性評価法を開発し、中・長期使用時の曝露riskを予測する」事を目的とした。
2016年度は、昨年度に引き続き、①極新鮮肝Bankの構築、②誘導性肝臓様細胞（iHep）を用いた感受性評価法の開発、③野生げっ歯類に注目したフィールドサーベイランス、④家禽（ニワトリ）、魚類（アフリカナマズ）、両生類（ゼノパス）を用いたin vivo投与試験を実施した。
①極新鮮肝Bankの構築では、昨年度から猛禽類を含む希少鳥類約30サンプル、ネコ科動物を含む哺乳類約20サンプルの採取を行った。②iHepの構築では、トド、アザラシ、ライオン、ネコ、ウサギ等の皮膚線維芽細胞の培養を行い、ストックの作成を行った。2017年度にはレンチウィルスベクターを用いた誘導を実施する。③野生げっ歯類をターゲットにフィールドサーベイランスを実施した結果、特に副腎や脾臓の肥大が顕著に観察され、DDTのターゲット器官である可能性が示唆された。④各種動物に投与実験を行い、毒性試験を実施した。

Current Status of Research Progress

Reason

極新鮮肝Bankの構築は、昨年度に引き続き順調に試料が集まっている。得られた試料から、特にPhase I (P450)、Phase II(UGT)を中心に、トキシコカイネティックス解析を順次行ったおり、大きな種差も観察で来ている。また、2016年度は昨年度は実施出来なかったフィールドサーベイランスを南アフリカ共和国クワズルナタールで実施した。歩哨動物として、ラットに注目し毒性学的解析を実施した結果、副腎や脾臓の肥大が観察され、DDTやピレスロイドによる毒性の可能性が示唆された。当該サンプルは現在解析中であり、2017年度の課題の一つである。
一方、野生動物の皮膚線維芽細胞から肝臓様細胞(iHep)の誘導であるが、皮膚線維芽細胞の採取は順調に進んでいるが、iHepへの誘導効率が未だ低く、更なる改良が必要である。また、当初計画していた野生動物の各種核内受容体、結合タンパク質、トランスポーターのクローニングについては2017年度に精力的に取り込む予定である。

Strategy for Future Research Activity

2016年度のフィールドサーベイランスで観察されたラットにおける副腎・脾臓の肥大は毒性学的に非常に興味深く、2017年度はトランスクリプトームやメタボローム解析等、様々な手法を用い、その毒性影響評価を実施する。また、2017年度も引き続きラットに注目したフィールドサーベイランスを継続する。また、極新鮮肝Bankの構築のための試料採集も継続して実施する。

iHepは誘導効率を上げるため、導入する因子の検討を行う。また、計画通り3次元培養を行うための準備も実施する。

一方、各動物のADMEを推定するにあたり、各種核内受容体、化学物質結合タンパク質およびトランスポーターのクローニングは重要であるため、当初予定通り進めていく。
