Outline of Annual Research Achievements

国内では虚血性心疾患,脳血管疾患などの動脈硬化を起因とする死亡原因が26%程度占めており,早期から心血管疾患リスクを軽減させるかが国内外ともに重要な課題となっている。近年,性ステロイドホルモンの新たな役割として動脈硬化との関連性が報告されているが,運動や食事による性ステロイドホルモン分泌の増大が動脈硬化改善に影響するのか否か,また,効果機序に関しては明らかでない。本研究は,運動により増大する血中性ステロイドホルモンが動脈硬化改善効果に貢献しているのかを性ステロイドホルモン合成酵素の合成阻害剤を用いて直接的な影響について検討することを目的とした。動脈硬化度が増加する肥満モデルラットとしてOLETFを用いて、トレッドミル運動25m/分スピード,60分間/日,週5日で8週間実施、運動トレーニング期間中、エストロゲンあるいは活性型テストステロンの合成酵素阻害剤をオスモティックミニポンプ法にて持続投与した。エストロゲンの合成酵素阻害剤をトレーニング期間中に投与した結果、血中エストロゲン濃度は有意に低下し、活性型テストステロンの合成酵素阻害剤をトレーニング期間中に投与した結果、活性型テストステロン濃度は有意に低下していたため、阻害剤による性ステロイドホルモン分泌低下モデルを作成することができた。また、肥満モデルラットの有酸素性トレーニングにより、動脈硬化度の指標である頸動脈-大腿動脈間の脈波伝搬速度(cfPWV)は有意に低下し、動脈血管のNO合成酵素のリン酸化発現も運動により有意に増大していた。一方、有酸素性トレーニング期間中、エストロゲン合成酵素阻害剤を投与したラットでは運動効果を減弱させることはなく、活性型テストステロンの合成酵素阻害剤を投与したラットでは運動効果に影響する可能性を示した。
