情動を制御するBDNFの直接効果の解明

Outline of Annual Research Achievements

BDNF遺伝子は9つの転写開始部位を持ち、9つのmRNA isoformを持つ。全脳BDNFノックダウンマウスとして2種類のマウスを樹立したところ、exon 4にSTOP cassetteをノックインしたマウス(BDNF STOP knockin)は、exon 1,2,3由来のmRNAの発現が消失し、exon 4を中心にtet systemで遺伝子発現を操作したマウス(Actin-tTS::BDNF tetO knockin)はexon 4,5,6,7由来のmRNAの発現が消失していることがわかった。いずれのマウスも、totalのBDNF mRNAの発現が約半分になっているが、前者は神経細胞由来のBDNF mRNAが抑制され、後者は、神経およびグリア細胞由来のBDNF mRNAが抑制されていることが期待された。前者は体重が少なく、後者は体重が増える傾向にあった。うつ・不安行動については、十分な個体数を使った解析はまだ出来ていない。
BDNF tetO knockinとdopamine transporter-tTAマウスを交配して得られるダブルトランスジェニックマウスでは、側坐核にBDNFが過剰発現する。このマウスを生後からドキシサイクリン混餌で飼育し、オペラント行動の獲得後から普通食にし、BDNFの過剰発現を開始した。BDNF発現操作前後で、意欲、注意、固執、衝動、食欲に変化は見られなかった。以上の結果は、今後、情動の制御における側坐核BDNFの役割を考える上で基礎データとなる。

性質の異なる全脳BDNFノックダウンマウスが幸運にも2つ樹立することが出来たが、一方で解析することが倍になった。このため、全脳BDNFノックアウトマウスの生化学的実験に時間をより多く割いている。行動解析にも倍の時間がかかる。

Strategy for Future Research Activity

性質の異なる全脳BDNFノックダウンマウスの行動解析を精力的に行っていく。
