Outline of Annual Research Achievements

温度制御システムの開発:好熱菌由来のタンパク質の機能動態やタンパク質間相互作用の温度依存性を調べるために、昇温・温度制御システムの開発を進めてきた。前年度までに完成した昇温システムの実用化を目指した。イメージング中に溶液温度をPCで設定し、フィードバック制御により溶液温度を1℃以内で維持できるようになった。また、昇温による溶液の蒸発で溶液容量が減少するのを防ぐため、あらかじめ測定された溶液の減少速度に対応して溶液が注入補填されるようにした。これにより長時間にわたって高温で観察ができるようになった。41度で固相から液相に相転移する脂質二重膜(DPPC)を用いて高温観察の評価を行い、相転移温度以上で膜の流動性が増加し脂質二重膜の形状が変化する様子を確認でき、43℃でも長時間に渡って安定にイメージング出来ることを確認した。
FliIの観察:温度制御システムを使って、べん毛の輸送タンパク質の一部であるFliIの観察を行った。溶液温度を38℃に設定しATP存在下でモノマーの観察を行ったところ、FliIモノマーが六量体のリング状オリゴマーを形成していく過程を観察できた。さらに、FliI六量体がリング状構造から、リング中心孔が塞がった異なるオリゴマー状態へ構造遷移する様子も観察できた。
KaiCの観察:シアノバクテリアの概日周期タンパク質であるKaiシステムの温度補償性に関する研究に温度制御システムを応用し、KaiAとKaiCの動的相互作用が温度によってどのように変化するか調べた。室温から28℃付近まではKaiAとKaiCの解離および会合の時定数は大きく変化せず、29℃以上になるとKaiAがKaiCに結合してから解離するまでの時間が短くなるとともに、解離してから結合するまでの時間も短くなり、KaiAとKaiCの積算結合時間は温度に依存せず一定になる可能性が示された。
