細胞機能を制御するDNAナノ構造体分子システムの開発

Outline of Annual Research Achievements

本年度は、人工的な受容体システムを開発するため、十字型のDNAオリガミ構造体を用いて2次元格子構造を構築し、脂質二重膜上での自己集合化の方法とその形成過程の可視化を行った。次に、十字構造体に針状構造体を中心に導入することで、脂質二重膜を貫通する格子状集合体と作成しリポソームと相互作用する系を構築した。マイカ上にDOPCによって脂質2重膜を作成し、この上に十字型のDNAオリガミ構造体を静電的に吸着した。脂質二重膜上では十字型のオリガミ構造体は十字構造の末端のπ-π相互作用によってモノマー同士が結合し、格子構造を形成した。特に、オリガミ構造体の溶液中の濃度が高い状態では、数マイクロメーターの格子構造を形成することを見出し、脂質二重膜上での流動性が拡張した格子構造を作るうえで重要であることがわかった。
作成した系を用いて、十字構造体に針状構造体を中心に導入することで、脂質二重膜を貫通する格子状集合体と作成し、脂質二重膜からなるリポソームと相互作用する系を構築した。十字構造の中心に4本の2本鎖DNAが結合した、剛直な20 nmの針状構造体を用いて中心を貫通するように設計した。この構造体を自己集合させると十字構造の中心に高さのある針状構造物が導入されているのがAFMから確認できた。これを用いて相補的に結合する2種類の十字型構造体を作成した。これらをアセンブルすると20個程度のモノマーが含まれた格子集合体の形成が見られた。これらの構造体をもつリポソームに加えると、その表面に結合することが蛍光顕微鏡観察から確認できた。

当初計画した脂質膜上でのDNAナノ構造体のアセンブリーの構築とその集合過程の可視化を行えた。脂質膜を貫通するDNAナノ構造体の構築も行えた。

Strategy for Future Research Activity

本年度は、作成したDNAナノ構造体を使ってリポソームとの相互作用とリポソーム内部への分子のデリバリーを行う。また、細胞との相互作用と細胞機能の制御の誘導を行える分子システムを構築する。
