細胞機能を制御するDNAナノ構造体分子システムの開発

Outline of Annual Research Achievements

本年度は、前年度から引き続き、人工的な受容体システムを開発するため、十字型のDNAオリガミ構造体を用いて、脂質二重膜との相互作用、特にリポソームと相互作用する系を構築した。十字構造体の中心に針状DNA構造体を導入することで、リポソームの脂質二重膜を貫通し、リポソーム内部に導入した生化学反応系を動かす系の構築を主に行った。
DNAオリガミ法を用いて作成した十字型構造の中心に4本の2本鎖DNAが結合した、剛直な40 nmの針状構造体を導入し、これを用いて脂質二重膜を貫通するように設計した。構造体を自己集合すると十字型構造の中心に高さのある針状構造物が導入されているのがAFMによって確認できた。次に、作成した十字型構造体をあらかじめ用意したDOPCからなるリポソームとインキュベートすることで、リポソーム表面に十字型構造体が結合することが共焦点蛍光顕微鏡観察から確認できた。さらに、針状構造の先端にDNA増幅反応を誘導するDNA鎖を結合し、そのDNA鎖が脂質膜を貫通し、DNA増幅系を導入したリポソーム内部で反応を開始できるか実験を行った。その結果、針状構造の先に反応を誘導するDNA鎖を導入した十字型構造体をリポソームに結合させることで、リポソーム内部のDNA増幅系が動作することが分かった。一方で、反応を誘導するDNA鎖がない針状構造のみでは増幅が見られないことから、針状構造部分が脂質膜を貫通することが示唆され、反応に必要なDNA鎖をリポソーム内部に送り込み、ターゲットとしたDNA増幅反応を開始できることが分かった。

Reason

当初の計画通りに、針状構造を導入した十字型DNA構造体の構築とリポソームとの相互作用の観察、及びリポソーム内部の導入した反応系の誘導を今回開発した十字型DNA構造体を使って行える系を作成できた。

Strategy for Future Research Activity

次年度は、外部環境に応答して動作するDNA構造体を開発し、リポソーム内部への分子のデリバリーと反応系の誘導を行う。また、細胞との相互作用と細胞機能の制御の誘導を行える分子システムを構築する。
