Outline of Annual Research Achievements

申請者が確立した、宿主ミトコンドリアへ輸送されたと思われるトキソプラズマのタンパク質リストに挙げられたタンパク質遺伝子にタグを付けて強制発現させることでミトコンドリアへの輸送を確認したところ、７つのタンパク質が寄生胞膜に局在し、宿主ミトコンドリアとの相互作用が示唆された。一方、このスクリーニングの過程で、強制発現させる遺伝子によってリクルートされるミトコンドリアの数が変化することに気づいた。つまり、タグ融合遺伝子の強制発現により原虫のミトコンドリアリクルート能力を亢進させる遺伝子群の存在が示唆できた。これらの遺伝子群はORF（Organelle Ricruitment Factor）の有力候補となるので、まず強制発現株によるオルガネラのリクルート能力の評価を行った。その結果、前述の7つの遺伝子のうち、2つが統計学的な有意差を持って、１つが有意差はないものの明らかにリクルート能力が亢進していた。現在、より詳細なリクルート能力の評価と、3つの遺伝子のノックアウトを試みている。

Reason

スクリーニングの過程で、タグ融合遺伝子の強制発現により原虫のミトコンドリアリクルート能力を亢進させる遺伝子群の存在が明らかとなったことは想定外であったが、逆にORF候補の絞り込みが容易になったことから、概ね順調に進展しているものと判断している。

Strategy for Future Research Activity

前述の通り、現在より詳細なリクルート能力の評価と、3つの遺伝子のノックアウトを試みている。これらの解析により、３つのORF候補遺伝子のオルガネラリクルートへの関与の実態が明らかにできるものと考えている。
