Outline of Annual Research Achievements

本研究では、CKDにおける低酸素による病態進行機転に関する研究を発展・新展開させ、CKDの病態生理についてエネルギー代謝とエピジェネティックな制御機構の観点から理解を深め、腎臓における低酸素応答の詳細を解析することを目的とした。
CKDにおける末期腎不全への final common pathway の主座となる尿細管細胞および酸素分圧の変化の影響に直接曝される血管内皮細胞の低酸素状態における遺伝子発現とエピジェネティックな変化を調べるため、培養細胞である HK-2、RPTEC、HUVEC などを用いて ChIP-Seq、RNA-Seqなどを行った。その結果、低酸素状態の血管内皮細胞には HIF-1alpha と Smad3 が同時に結合して様々な線維化促進分子の発現を調節することが分かった。また、低酸素状態の尿細管細胞では DARS-AS1 という新規 long non-coding RNA が HIF 依存性に発現し、エピジェネティックに細胞の低酸素におけるアポトーシスに対する抵抗性を惹起していることが分かった。更に、動物モデルにおける急性の虚血再灌流障害後の長期的な腎線維化を AKI-to-CKD transition のモデルとして確立し、これにヒストン修飾を調節する薬剤を投与することにより腎臓の線維化が改善することを示し、エピジェネティックな機序による hypoxic memory の重要性とそれをターゲットとした治療の有効性を示した。
