Elucidation of molecular architecture of neuromuscular junction and dissection of molecular pathomechanisms of congenital myasthenic syndromes

Outline of Annual Research Achievements

Wntリガンドが胎生期におけるアセチルコリン受容体(AChR)のクラスタリングに重要であることが知られてきたがそのシグナル伝達に関わる分子は十分に解明されていない。マウス脊髄前角細胞(SMN)のレーザーキャプチャーマイクロダイセクションのマイクロアレイならびにRNA-seqによる網羅的な遺伝子発現解析により、Wntシグナルを制御する細胞外分泌タンパクRspo2が脊髄前角細胞に特異的に発現することを見出した。Rspo2をSMN特異的にレスキューするvAChT-Rspo2 x Rspo2-/-マウス、ならびに、Rspo2を骨格筋特異的にレスキューするMCK-Rspo2 x Rspo2-/-マウスを作出して神経筋接合部(NMJ)構築の解析を行った。SMNに発現させたRspo2は、異常に拡大したAChR領域・NMJ超微細構造の約半数のパラメータ・SMNと骨格筋の遺伝子発現を正常化させた。一方、骨格筋特異的に発現させたRspo2は、NMJ超微細構造のすべてのパラメータを改善させたが、光学顕微鏡レベルのAChRクラスタリング・NMJ形成、ならびに、遺伝子発現には効果を認めなかった。SMN由来のRspo2がAChRクラスタリングとNMJ構築に重要な役割を担っていることが示された。分泌タンパク質のためNMJにおけるRspo2の起源の同定は困難であるが、骨格筋由来Rspo2にも一定の効果が認められた。
さらに、GFPT1遺伝子変異を有する本邦先天性筋無力症候群患者から樹立したiPS細胞を用いて病態分子機構の解明研究を開始した。
