Outline of Annual Research Achievements

免疫療法の標的抗原の探索
1. リコンビナント蛋白質を用いた標的抗原の探索：大腸菌で作成した変異型（Val30Met）TTRを酸性条件下でインキュベーションしTTRを変性・アミロイド線維化させる。本過程で様々な形態の変性TTR、オリゴマーなどが形成されるため、HPLC（ゲル濾過カラム）を用いて分子サイズにより分取した。さらに異なった症状のFAP(劇症型、髄膜型、心臓型など)の原因となる各種の変異体TTR（Leu55Pro、Tyr114Cys、Ser50Ileなど）を用いて同様の検討を行った。
2. FAP患者組織から抽出・分離・精製した抗原：FAP患者の剖検凍結組織をホモジネートしPras法でアミロイド線維を抽出した。この過程で得られる可溶性の分画もストックし、HPLC法で分取後にTTRオリゴマーの検出した。得られた各種のTTR形態を精製後に抗原として用いた。解析の結果、TTR115-124はTTRがアミロイドになる折に確かにcryptic epitopeになることが確認されたが、加えて、TTR88-96も同様の抗原になることが新たに判明した。
3. iPS細胞と組み合わせたin vitroの研究
アミロイドを形成するTTR81-127 を細胞にトランスフェクトし、iPS細胞+TTR89-96抗体およびTTR115-124 抗体を添加し、アミロイド抑制効果を見たところ、顕著な抑制効果が得られた。

Reason

TR115-124はTTRがアミロイドになる折に確かにcryptic epitopeになることが確認されたが、加えて、化血研との共同研究に加えて、アメリカのベンチャー企業であるProthena社とも共同研究をしており、今年度予定しているiPS―マクロファージの研究を行いながら、FAP患者に対する応用を目指して、確実に前に進んでいる。前臨床試験に入る具体的な段階に入っている。
現在までのところ要所の予想通りに研究は進展しており、あらたな研究の変更はない。

Strategy for Future Research Activity

平成29年度は前年度の研究を継続して実施し不足分を補う。治療手法を組み合わせた複合的な免疫療法の効果検証を行う。また、モデル動物に対する副作用が増強されないか特に注意して解析する。また細胞療法の開発の研究も開始する。具体的には以下の通りである。
1. 健常人およびFAP患者よりiPS細胞を樹立：健常人とFAP患者の末梢血リンパ球よりiPS細胞を樹立する（すでに複数の患者より疾患特異的iPS細胞を樹立している）。
2. マクロファージへ分化させ、各抗原の貪食効果を解析：健常人由来とFAP患者由来のiPS細胞からマクロファージへ分化させる。双方のiPS-MΦに（A）で得られた標的抗原を投与し、貪食能や反応性（サイトカイン産生）などに違いがあるか検証する。
3. モデル動物での効果検討：健常人由来とFAP患者由来のiPS-MΦを（A）で得られた標的抗原と共にインキュベーションした後にモデルラットへ投与し、TTR沈着量の変化などを検証する。
4. 副作用の確認：iPS-MΦの投与に伴う、副作用（生体での炎症反応）の確認をラットやウサギを用いて実施する。
