Outline of Annual Research Achievements

ヒトiPS細胞から神経堤を介して誘導したMSC(iNCMSC)を作成し、性状分析を施行した。
神経堤細胞を血清培地で培養後1-2週以内にMSCマーカー(CD44, 73, 105)陽性かつ神経堤マーカー(CD271)陰性の細胞集団が再現性高く得られることがわかり、さらに多能性マーカー(TRA-1-60, Oct3/4, Nanog, Sox2, Lin28a)の発現の消失および減少が確認された。
iNCMSは、In vitroにおいて、骨分化誘導により早期でのALP発現上昇、その後の石灰化基質の沈着を認め、骨分化関連遺伝子の発現上昇も認めた。また、軟骨分化試験では、サフラニンO陽性、2型コラーゲンリッチな硝子軟骨様組織が形成され、培養日数に伴う軟骨分化の関連遺伝子、GAG量の上昇を認め、軟骨様の組織を形成することも確認された。しかし、iNCMSCを高密度培養することで得られる人工組織(iNCMSC-TEC)を免疫不全ラットの軟骨欠損部に移植すると、欠損部に線維組織が充填され、軟骨下骨のリモデリングが著しく阻害されていた。残存細胞は継時的に追跡すると消失していき、Ki67などの増殖マーカーの発現も認められなかったことから腫瘍形成などは認められなかった。結果として、iNCMSCはin vitroでは高い軟骨分化能を示したことから軟骨治療において有用な治療オプションにはなりえるが、in vivoでの結果を踏まえ、臨床応用までにはさらなる検討が必要であると考える。
