海底下に潜む未知の生命－多面的アプローチで明らかにする地球深部微生物の実態

Outline of Annual Research Achievements

研究初年度である平成27年度は、研究実施計画として予定していた通り複数の掘削研究航海で取得した試料についての安定同位体基質を添加した培養を継続したほか、北大西洋中央海嶺付近、アトランティス岩体において実施された国際深海科学掘削計画（IODP）第357次航海に乗船研究者として参画し、新たな海底下岩石試料を取得した。試料としては、海底面から掘削を行って得られた海底下岩石試料のほか、試料の汚染源となりうる掘削時に掘削ビットを冷却する目的で循環される掘削用水、及び試料を船上に揚収するまでに暴露される可能性のある海水をそれぞれ取得した。下船後、海底下岩石試料についてはまず速やかに医療用CTスキャン装置によって試料の三次元構造を明らかにした。その後、掘削用水によって汚染が疑われる外周部をクリーンブースに設置したバンドソーシステムで除去した。バンドソーの切断にあたっては、試料ステージを冷却して行い、不必要な試料の温度上昇が起こらないように配慮して行った。その後、内側部分をさらにバーナーで火炎滅菌してから嫌気グローブボックス内で岩石試料を粉砕し、バイアル瓶に取り分け、各種安定同位体基質を添加して培養を開始した。培養中のバイアル瓶は、微生物による基質転換反応の兆候をとらえるため、ヘッドスペース中の二酸化炭素の安定同位体比を定期的に調べている。
環境試料からの細胞取り出し（分離）も、これまでに培養した試料について順次進めており、NanoSIMSによる分析のための試料準備を進めている。

上記、研究実績の概要に記載した通り、平成27年度の研究として実施を予定していた項目についてはおおむね達成することができた。

Strategy for Future Research Activity

しかし、予想通り、IODP航海乗船とその試料処理に3か月程度の時間が必要で、NanoSIMSによる分析を進めることが出来ていないが、ヘッドスペースの二酸化炭素などの同位体比分析を通じて活性の高いものをあらかじめピックアップするなどして効率化を加えながら平成28年度の研究を実施したい。
