HLA imputation法を用いた自己免疫疾患のバイオマーカーの同定

Outline of Annual Research Achievements

ヒト主要組織適合性複合体(MHC)領域内のHLA遺伝子配列の個人差は、多数の自己免疫疾患の遺伝的背景の大半を説明する。しかし原因HLA遺伝子多型の同定は、HLA遺伝子の複雑な構造や、高額なアッセイ費用、データ解析手法の難解さ、等の事情により遅れていた。本年度の研究においては、ゲノムワイド関連解析データにおけるHLA遺伝子多型を遺伝統計解析手法によりコンピューター上で高精度に決定し、追加費用無しでの網羅的解析を可能にする、「HLA imputation法」の日本人集団への適用拡大を行った。日本人集団900名を対象に、MHC領域内における高密度一塩基多型(SNP)および古典的HLA遺伝子配列(クラスI HLA遺伝子:HLA-A/B/C、クラスII HLA遺伝子:HLA-DRB1/DQB1/DPA1/DPB1)を実験的に決定し、日本人集団におけるHLA imputation法実施時の参照配列を作成した。自己免疫疾患の1つであるバセドウ病の大規模ゲノムワイド関連解析に対してHLA imputation法を適用し、複数のHLA遺伝子(HLA-DPB1/A/B/DRB1)におけるアミノ酸配列多型が発症に寄与していることを明らかにした。また、新学術領域ゲノム支援班の支援対象課題に採択された。
