| Outline of Annual Research Achievements |
難治性癌の代表である膵癌・胆道癌の病態には、癌細胞の上皮間葉転換 (epithelial mesenchymal transition、EMT) を伴う浸潤性増殖、および脈管侵襲を介した転移が関与している。この癌細胞の上皮間葉転換には癌細胞と癌関連線維芽細胞との関連が重要である。胆管癌の外科切除標本を用いた検討では、原発巣、リンパ節転移巣において癌関連線維芽細胞の多い症例で優位に予後不良となることを明らかにした。また、胆管癌細胞株とヒト間葉系細胞株との共培養系モデルにおいて、癌細胞の形態変化、免疫染色で癌細胞におけるvimentin の発現上昇、E-cadherin の発現低下といった、上皮間葉転換が生じることを明らかにした。また同環境下における時計遺伝子の関与を検討したところ、共培養を施行した胆管癌細胞において、DEC1の発現が上昇していることを、RT-PCT を用いて明らかにし、時計遺伝子の制御のもと、癌微小環境が形成されることが示唆された。さらにより生体に近い癌微小環境モデルの作製のため、リンパ管組織を含む三次元培養モデルにおいてリンパ管に癌細胞が浸潤する組織像を得ることができた。
