Outline of Annual Research Achievements

悪性中皮腫の約80%において、アスベスト暴露が発がんの原因といわれているが、日本ではアスベストの規制が遅れたこともあり、今後の罹患者数増加が予想されている。一方、悪性中皮腫に対する治療については、手術と薬物療法が選択肢となるが、手術の有効性は確立されていない。また、薬物療法においても効果は十分ではない。そのため、有効な新規治療法の開発に対する期待は非常に大きい。我々はこれまでに悪性胸膜中皮腫症例の一部においてVHL変異を有する症例が存在し、新規治療標的となり得る可能性があることを報告した。本研究においては細胞株を用いて、VHL変異を標的とする治療開発へ向けた基盤研究を実施する。
初年度にVHL変異悪性胸膜中皮腫細胞株NCI-H28およびVHL野生型悪性中皮腫細胞株であるNCI-H2052とMSTO-211Hを用いて基礎検討を実施した。VHL変異によりHIF-1αの恒常的な高発現を認めるとともに、HIF-1α阻害剤によるアポトーシス誘導による増殖抑制効果が確認された。これらの結果から、VHL変異悪性胸膜中皮腫細胞株の増殖がVHL変異に起因するシグナル伝達系に依存することを示すとともに、HIF-1α阻害剤に対して特異的感受性を示すことを確認した。さらなる検討のために、VHL無発現細胞株である786-O細胞株にレトロウイルスベクターを用いて野生型VHL及び変異型VHLの導入を実施行い、VHL強制発現細胞株モデルの樹立を行った、。現在はこれらの細胞株において、HIF-1α阻害剤を含めた薬剤感受性に対する影響についての検討を進めている。加えて、臨床検体を用いてのさらなる検討のために次世代シークエンシングの実施を進めている。研究計画については既に当院倫理委員会から承認を得ており、現在検体の収集及び解析を継続して進めている。
