Development and mechanism analysis of small molecules affecting tumor-stromal cell interactions

Outline of Annual Research Achievements

平成29年度は、これまでに引き続き、蛍光タンパクGFP遺伝子を安定的に導入したヒトがん細胞株と対応する臓器由来のヒト間質細胞との共培養系を用いて活性物質の探索を行った。その結果、肺がんおよび膵がんの共培養実験系で、がん細胞および間質細胞のみの培養に比べて共培養した時にがん細胞の増殖を強く阻害する活性を放線菌やカビの培養液に見出した。活性物質が微量であったため、活性物質を生産する微生物を大量培養することによって構造解析に必要なサンプル量確保のため活性物質の精製を継続している。一方、これまでの検討から活性を見出した8-azaguanine (8AG)については、作用機構の解析を行った。肺がん細胞６株について肺間質細胞との共培養系で8AGの活性を検討した結果、NCI-H460およびNCI-H522細胞の増殖を共培養下で強く阻害することが分かった。8AGを処理した間質細胞の培養上清では活性の増強は認められず、がん細胞と間質細胞の接触が活性に重要であることが示唆された。一方、同じプリン類縁体である6-thioguanineおよび6-mercaptopurineでは同様な活性は見られなかったことから、8AGに特異的な作用機構が存在すると考えられた。そこで、感受性を示した肺がん細胞株２株で他の細胞よりも特徴的に発現している分子をアレイデータから抽出した。現在、候補遺伝子産物と8AGの作用との関連について検討を行っている。
